Abstract
A research had been done of bioactivity of Chromohalobacter sp bacteria from Callyspongia sp sponge toward of Salmonella typhii and Staphylococcus aureus. The process of this research include: fermentation pure culture, extraction and has done by determining of ability an inhibit the growing of the tested bacteria was done by KirbyBaeur difusion method to get the MIC value in concentration of 0,1, 0,5, and 1%. TLC-biaoutografi test have been done to get the compound which had antimicrobial activity. The goal was to find a potential antibacterial ethyl acetate extract of bacterial Chromohalobacter sp symbion Callyspongia sp sponge on the growth of Salmonella typhi bacteria test and Staphylococcus aureus and compares to the chloroform extract Callyspongia sp sponges and determining which groups are antibacterial compounds from these two extracts. The results of determination of the resistance from the ethyl acetate extract Chromohalobacter sp shows that this extract is a narrow-spectrum antimicrobial that inhibits the growth of only gram-positive bacteria (Staphylococcus aureus) and are bacteriostatic. While from chloroform extract Callyspongia sp is a broad-spectrum antimicrobial that inhibits the growth of gram-positive bacteria (Staphylococcus aureus) and gram-negative (Salmonella typhi) and are bacteriostatic. The separation of compounds with TLC performed used dilution of eluent Chloroform: Methanol (10: 1) result are 4 stain with Rf values different from the ethyl acetate extract of Chromohalobacter sp those are 0.86; 0.51; 0.46 and 0.40. While the chloroform extract Callyspongia sp result are 3 stain with different Rf value of 0.87; 0.51 and 0.81. Testing the TLCBioautographic Contacts method showed 1 stain active than ethyl acetate extract of the Chomohalobacter sp Rf values of 0.86 and group identification showed alkaloid compound. While from chloroform ekstrak Callyspongia sp showed 3 active stain with Rf value of 0.87 is identified as a group of steroid compounds, Rf 0.51 terpenoid class of compounds, and Rf 0.81 group alkaloid compounds. This study showed that the ethyl acetate extract Chromohalobacter sp able to produce the same compounds with chloroform extracts Callyspongia sp are both capable produced alkaloid compounds, and both groups showed antibacterial activity.
